The author review in details the mechanism of action of neuroleptic drugs. The potent ones are tertiary amines with difinite chemical and physiological properties to inhibit the permeabulity of lipid-water interphases, be formation of momolayer at the clefts of dopaminergic neurones. Since potent neuroleptics and GABA have similar structures, the former ones will occupy GABS receptors where they display GABA-like action in prevention of membrane polarising. Conseguently, either liberated dopamine or injected amphetamine is unable to reach dopamine receptors; by which means, the nerve impulse is blocked. In brief, the sites of actions of neuroleptics are:- 1. To inhibit vomiting by acting at the dopaminergic emetic chemo-receptor or trigger zone in area cinerea at the floor of the fpurth ventricle 2. To prolong reaction time by acting at the nigro-striatum dopaminergic system. 3. To inhibit the action of amphetamine, producing stereotyped movements by acting at the nigrostriatum dopaminergic system, staring at A9 area or parscompacta of locus niger.
