The serotonin transporter-linked promoter region (5-HTTLPR) polymorphism occurred by insertion/deletion is evident to result in three different length alleles namely S,L as predominant variant alleles and XL as a rare one. These three alleles have 14, 16 and 18 or 20 repetitive DNA sequences, respectively and have been shown to associate with certain neuropstchiatric diseases. The 5-HTTLPR polymorphism has been demonstrated to significantly depend on ethnic groups, and it today becomes of great importance for pharmacogenomics in psychiatry. However, the 5-HTTLPR genotype distribution and allele frequency in Thai population have not been elucidated. Variable number of tandem repeat polymorphisms (VNTR) including this particular one are frequently analyzed by conventional polymerase chain reaction (PCR), but it is difficult for the 5-HTTLPR polymorphism genotyping due to high GC-rich genomic DNA templates. Therefore, the use of high-cost chemicals such as 7-deaza-dGTP in PCR reactions has been noted to minimize the PCR dilemma. Herein, we successfully developed a new PCR approach called touchdown (TD) PCR, a versatile one-step procedure by which primers would avoid amplifying nonspecific sequence without using the deaza-dGTP. The protocol relied on incremental annealing temperature decreases in progressive cycles designed to bracket the melting temperature ™ of the reaction. The genotyping result could be satisfactorily validated using a Taq polymerase manufactured for GC-rich templates. Our novel finding will be of great benefit in genotyping ine 5-HTTLPR polymorphism in laboratories.
