Rapid urinary methamphetamine detection kit was developed based on the technique of immunochromatography. The kit was prepared by fixing nitrocellulose membrane with methamphetamine conjugated with bovine serum albumin (MA-BSA), and anti-mouse immunoglobulin G (Anti-mouse IgG) at the test (T) and control (C) lines, respectively, whereas the anti-methamphetamine monoclonal antibody labelled with colloidal gold was fixed at the conjugate release pad (CRP). These revealed the sensitivity against methamphetamine as 1,000 ng/ml of urine sample, of which the negative result was shown as pinkish-purple lines at both T-and C-position, while the positivity was determined as only one line at C-position. These could be detected within 5 minutes, and the observation should be done no longer than 8 minutes. As cross-reactivity study, 35 analogous compounds were determined, and 3,4-methy-lenedioxymethamphetamine (MDMA) or ecstasy and other drugs crossed react with the test. For the stability, the kit was stable for 2 weeks at 60C, 12 weeks at 45C 24 weeks at 37C. It was concluded that the kit should be stable up to 2 years at room temperature. Moreover, the kit was performed laboratory evaluation by comparing with 1,461 urine sample confirmed with thin layer chromatography (TLC) which showed the clinical sensitivity and specificity as 97.0% and 98.7%, respectively. Comparative study between DMSc kit and commercial kit demonstrated the sensitivity 96.8% and 94.9% respectively with the specificity at 100% for both kits. In addition, field evaluation with 5,882 urine samples which were positive 4,521 samples and negative 1,361 samples, the result gave the sensitivity and the specificity 99.6% and 99.5% respectively.
